Shuttle, dark (box) experiment method

The avoidance experiment was established by using the animal's good darkness (light and dark) and fear and memory of aversive stimuli (such as electric shock). So for the experimenter (rather than for the animal), the avoidance experiment is technically similar to suggesting the associated conditional fear. The stimulus used was a mild foot shock, and the response was that the animal escaped from the area that was previously stimulated by electric shock. According to the escape mode of animals, they are divided into active avoidance and passive avoidance . The former requires animals to actively escape from a box with aversive stimuli; the latter requires animals to curb themselves without entering a box with aversive stimuli. According to the different instruments and equipment used, it can be divided into shuttle avoidance, platform avoidance, and Y- shaped labyrinth avoidance. Here are the main two avoidances.

First, the experimental equipment

(1) Shuttle avoidance experimental equipment Includes shuttle box, stimulus controller and computer. Here is the ENV-010MC shuttle box produced by Med Associates . It comprises about two equal size (20.3cm _* 15.9cm _* 21.3cm) compartment (compartment). One of the compartments is wrapped with a black cloth cover to form a black box (active avoidance can be used without a black box). The two compartments are connected by an arched door ( 8.9 cm _* 11.4 cm ), which can be closed or opened by the experimenter according to the experiment. The compartment floor is a stainless steel fence (diameter: rat 4.8 mm , mouse 3.175 mm ). The fence floor on the side of the black box is connected to the stimulator and can be given an electric shock; the side of the bright box is not connected to the stimulator, so no current flows.

(2) Ski jumping avoidance experimental device The middle door of the shuttle box is removed and replaced with a platform (which can be used with the Med Associates shuttle box) to be converted into a platform avoidance experimental device. Here is a platform evasion device from Coulboum Instruments, another US instrument company . It consists of a transparent plexiglass jumper of 15cm _* 25cm _* 10cm , a soundproof outer box ( 48cm _* 62cm _* 24cm ) and a pulse electrical stimulator. The fence at the bottom of the skid box is connected to the stimulator, and a 3.5 cm _* 3.5 cm _* 2.5 cm wooden platform is fixed in the center . During the experiment, put the jumper box into the soundproof box to prevent external interference.

Second, the experimental method

(A) a passive avoidance test in mice shuttle (mouse step-through passive avoidance)

1 . Single shock stimulation training

( 1 ) Put the mouse in the bright box, and after 10s , open the door between the light and dark boxes . Most strains of mice have a strong inquiry behavior, like dark evil. Therefore, the mouse will quickly enter the dark box.

( 2 ) Immediately after the mouse (four limbs) completely entered the black box, the middle door was closed and an electric shock was given. The shock intensity is the minimum current that causes the mouse to retract and vocalize. According to the mouse strain, the current range is 0.2~0.8mA (100V , 50~60Hz AC) for 1~2s .

( 3 ) Let the mice stay in the dark box for 10 s (so that the animal forms a correlation between the box and the shock) and return the mouse to the cage. Clean the operating box. Carry out training for the next animal.

2 . Memory retention test (retention test) After 24 hours of electrical stimulation training , the mice were returned to the clear box, and the middle door was opened, but no electric shock was given.
